Abstract : [Abstract] To prevent the extraction of lipids inside and outside an osteoblast during a double staining procedure for electron microscopy, some specimens which had been fixed with glutaraldehyde containing malachite green and osmium tetroxide were immersed for 2hr in the cacodylate buffer (pH9.0) containing 0.5% p-Phenylenediamine and 0.5% Imidazole immediately after postfixation. The stained sections showed blackened, well circumscribed lipid droplets similar to those in corresponding unstained sections. This staining procedure is considered to be useful in preventing lipid extraction during section staining.
